D-amino acid oxidase (DAAO, DAO, EC 1.4.3.3) is a typical flavin protease with flavin adenine (FAD) as a cofactor, which oxidizes the amino groups of D-amino acids to produce corresponding ketoacids and ammonia. The catalytic reaction is as follows:

D-amino acid + O2 → ketoacid + H2O2 + NH3

The D-amino acid oxidase combines with the coenzyme FAD and reacts with the D-amino acid. The dehydrogenation of D-amino acid becomes the corresponding sub amino acid and combines with the reductive FAD. This step is the only enzymatic reaction in the whole reaction. In the presence of molecular oxygen, the reductive fad is oxidized again and oxygen is converted into hydrogen peroxide; the sub amino acid is hydrolyzed into A-ketoacid and ammonia, However, when hydrogen peroxide is present in the body, A-ketoacids decarboxylate to produce corresponding acids.

D-amino acid oxidase has high stereoisomerism selectivity and broad spectrum for catalytic reaction substrate, which can be widely used in qualitative and quantitative analysis of D-amino acids, biosensor, production of L-amino acids and A-ketoacids.
